Gene mutation shown to cause leukemia and lymphedema

ScienceDaily (Sep. 5, 2011) ? Researchers have discovered a gene that when mutated can cause lymphedema (swollen limbs due to a failure of the lymph system), immune abnormalities, deafness and leukemia. The identification of the gene responsible for causing this rare combination of medical conditions, known as Emberger syndrome, could allow earlier identification and treatment of those at risk.

This study, which is published online in the journal Nature Genetics on Sept. 4, showed that it is caused by a mutation in the GATA2 gene.

Mutations in this gene have very recently been associated with the development of leukemia and immunodeficiency but had not previously been linked to lymphedema.

The study was led by a team at St George's, University of London and St George's Hospital. The researchers recruited eight patients with Emberger syndrome, two of whom had a family history of the condition. In collaboration with colleagues at Kings College London, they carried out Next Generation Sequencing (a new technique which can rapidly sequence all the genes in the human genome) on these patients to identify any genetic patterns that could provide clues about the disease. All eight patients had a mutation in the GATA2 gene.

Consultant geneticist, Dr Sahar Mansour, who led the study with Professors Steve Jeffery and Peter Mortimer, said: "Emberger syndrome affects only a handful of people in the UK. However, little is known about the development of both the lymphatic system or leukemia This gene obviously has a crucial role in both and is an important piece of the jigsaw puzzle. It may lead to a better understanding of why these problems develop and eventually target treatment for both."

The identification of a mutation in GATA2 adds to the increasing number of genes responsible for causing Primary Lymphedema. It is the fourth gene to be associated with the condition to be identified at St George's, which runs the UK's only specialist clinic for primary lymphedema.

Professor Mortimer who runs this specialist clinic said: "The genetic causes of lymphedema are important developments in the understanding of both cause and mechanism for lymphatic disorders which are a neglected area of clinical practice. Gene testing is already changing the means by which lymphedema is categorised and diagnosed. In the future it will lead to treatment through drugs acting on molecular targets."

Business Brief Japan| International Business News | Conversis | The ...

Conversis? business briefs are a series that give you a quick summary of the social, political, economic and cultural news that you need to be aware of if you?re doing business in a foreign country.

Here?s a snapshot of the information you get in a business brief.

Is Japan a good country to do business in now?

Perhaps, but only with local partners. .

Here?s why

Japan is the world?s third largest economy and arguably the most technologically advanced country in the world. It is a leader in scientific research, the manufacture of innovative technological products, robotics, health care and medical research. Japanese business culture remains distinctly unique and Western companies often find it difficult to succeed there. Although barriers to foreign investment have largely decreased in the past decade, strong local companies will compete fiercely against new entrants to their markets. Japanese customers can be very demanding and often have quite different tastes and needs to Westerners. This means that companies must redesign or redevelop products in order to succeed in Japan. Because of Japan?s size, substantial investment is necessary to enter the market, creating considerable risk.

Fast Facts

Population: 127 million
Language: Japanese
Religion: Shintoism, Buddhism; many people belong to both
Land area: 377,864 square km
Currency: Yen
Capital: Tokyo
Internet domain: .jp
International dialing code: +81
Innovation ranking[1]: 1 (out of 82 countries)
Digital economy ranking[1]: 16 (out of 70 countries)

Economic indicators: 2011

Real GDP growth: -0.9
GDP per capital (US$): $37,192.71

Economy

On 11 March 2011, Japan experienced its greatest disaster of the postwar era when an earthquake and tsunami devastated vast areas of eastern Japan and sparked a nuclear crisis. According to the OECD, the immediate impact of this has been to reduce output, although this is likely to be reversed in the second half of 2011 led by reconstruction efforts. However, deflationary pressures are likely to continue through 2012, with unemployment remaining above its pre-2008 financial crisis levels.

Reconstruction spending will be significant given the scale of the disaster, but with public debt exceeding 200% of GDP, economists and markets will be closely watching how the reconstruction is financed.

Politics

Japan has a constitutional monarchy with a parliamentary government. Legislative and executive powers are vested in the Diet (parliament), which is composed of the House of Councillors and the more powerful House of Representatives. The emperor serves as head of state and as a symbol of the Japanese nation and the unity of its people in a purely ceremonial role. The current Emperor, Akihito, succeeded his father, Hirohito, in 1989.

The prime minister serves as the head of government. The current prime minister, Naoto Kan, came to power in June 2010 following the resignation of Yukio Hatoyama after a controversy surrounding his failed election promise to move a US air base. Mr Kan is the fifth premier in four years.

Doing business

When conducting business in Japan, remember the following few guidelines:

  • Avoiding conflict and trouble is extremely important in Japan and diplomatic language is often used rather than the direct approach. In formal situations a direct ?no? is avoided.
  • Japanese have been raised to think of themselves as part of a group. Whether at a national, school or company level, Uchi-soto (?us and them?) applies. This works well in business, with manufacturers, suppliers and distributors working together in closely-knit groups called keiretsu.
  • The move to Westernisation is slow. You may find the more Westernised a Japanese business person is, the less they are respected by their Japanese colleagues.
  • In all top Japanese companies with offices outside Japan, the major decisions are made in Japan and the process is slow. They have an all-inclusive decision making process that can seem laboured to Westerners. When decisions are made, however, action follows very quickly.
  • The Japanese can set tough standards, but they can also offer help when asked. They build close relationships along the supply chain and, when trusted, will be happy for you to leverage it.

Berlin court rules far-right posters can remain (AP)

BERLIN ? A Berlin court ruled Wednesday that campaign posters for the far-right National Democratic Party with the slogan "step on the gas" may remain on display.

The placards is one of several the far-right party is using in an effort to attract votes for upcoming Berlin state elections. One shows caricatures of Muslim immigrants on a flying carpet with the slogan "have a nice flight home." Another bears the slogan "step on the gas," with a picture of the party's leader on a motorcycle.

Berlin's Kreuzberg district had ordered the posters taken down, saying they violated German incitement laws.

But the Berlin state court ruled Wednesday that the posters were protected under freedom of speech laws. It noted that although the "step on the gas" poster "could well be intended to arouse associations with Nazi atrocities," it could also have other meanings.

A U.S. group of Holocaust survivors expressed "horror and disbelief" at the ruling and called for the posters to be banned.

The party isn't expected to win any seats in the Sept. 18 vote.

Film "Shame" brings sex addiction tale to Venice (Reuters)

VENICE, Italy (Reuters) ? Up-and-coming Irish actor Michael Fassbender plays a sex addict in "Shame," a movie by British video artist Steve McQueen that is vying for the top prize at the Venice film festival.

It is the second lead role for Fassbender in a competition movie at this year's festival after his portrayal of psychoanalyst Carl Jung in David Cronenberg's "A Dangerous Method."

In Shame, the German-born Fassbender is Brandon, a handsome, 30-something executive living in New York whose only distraction from work is seducing women, masturbating at home or in the office and looking for sex on the Internet.

The tightly controlled rhythm of his life begins to fall apart when his needy, dysfunctional sister Sissy, played by Carey Mulligan, arrives for an unannounced visit.

Her presence, and her craving for Brandon's attention, disrupt his lonely existence even further, and his only way out seems to be wandering the streets at night in search of new sexual adventures.

Fassbender, whose portrayal of Brandon was warmly applauded after a press screening on Sunday, said taking part in the film's graphic sex scenes was not easy.

"Yes (it was) uncomfortable doing the sex scenes, you just have to jump and turn really," he said.

"The most important thing I guess is that everybody involved feels as comfortable as they can. And then just go for it so you don't have to do too many takes."

McQueen, whose debut film was the widely acclaimed "Hunger" about the last months of Irish Republican Army activist Bobby Sands in Belfast's Maze prison, said he saw similarities between the two films.

Hunger also starred Fassbender in the lead role.

"Clearly Hunger was a political film but Shame is also political. That one was about a prison in northern Ireland, this one it's about how someone's freedom can actually imprison them and they need an addiction in order to numb a pain, how our lives have been changed sexually by the Internet," he said.

"I love Brandon, he's trying and it's difficult. He's not so far away from most of us at the end of the day. He is not a bad person, I think the character is not at all repulsive, maybe unfamiliar but extremely recognisable."

The title Shame was chosen after interviews with sex addicts and their experiences in preparation for the film.

"The word shame came cropping up in those interviews," McQueen said.

His career began with film-related projects, he quickly branched out to include sculpture and still photography, and his work has been displayed at the Biennale of Art in Venice.

McQueen said he saw no big differences between his artworks and his feature films.

"There are no barriers between the two. Of course in one you're going to have a bit more narrative and the other less so, but the process is the same, it's work."

Asked why Mulligan had not come to Venice to present the movie, McQueen replied: "It's out of order. She should be here."
